The citric acid cycle enzyme aconitase is a member of a growing family of superoxide anion (O2.-) sensitive [4Fe—4S]-containing (de)- hydratases that has been implicated to be important sites of superoxide anion toxicity. The activity of aconitase is sensitive to inactivation by level of O2.- and is modulated by changes in O2.- levels in bacteria and mammalian cells. Thus, inactivation of aconitase can occur in response to oxygen radical generating compounds such as redox cycling agents, higher levels of oxygen or pathological conditions resulting in higher levels of O2.-. The assay is based on conversion of citrate to isocitrate by aconitase, and conversion of isocitrate to α-ketoglutarate by isocitric dehydrogenase. In this process, NADP+ is converted to NADPH, the absorbance of which is measured at 340 nm. The activity of aconitase is calculated as the rate of formation of NADPH and is expressed as milliunits of aconitase/mg protein.
