α-Ketoglutarate dehydrogenase complex is a multienzyme complex present in the mitochondria. This enzyme catalyzes the oxidative decarboxylation of α-Ketoglutarate (KG) to succinyl-CoA with concomitant production of NADH which is proportional to the activity of the enzyme. This enzyme has been shown to be inactivated by oxidative stress. Therefore, the activity of this enzyme is a likely target of oxidative inactivation. The assay procedure uses 1-mL assay reaction volume, and requires quartz cuvettes of 1-mL capacity and a spectrophotometer with kinetic function with temperature control. The assay can be performed with isolated mitochondria or whole cell or tissue lysate.
