Superoxide Dismutases (SODs), also known as manganese-dependent superoxide dismutase (MnSOD, SOD2), were initially discovered as indophenoloxidases (IPOs) which are endogenously-produced intracellular antioxidant enzymes that catalyze the conversion of superoxide radical anions produced from oxidative phosphorylation into hydrogen peroxide (H2O2) and further catalyzed to innocuous oxygen (O2) and water (H2O) by glutathione peroxidase and catalase. Broadly this serves as a defense mechanism against reactive oxygen species (ROS).

SOD2 is a homotetramer with a mass of 80 kDa and is a manganese-containing enzyme residing in the mitochondrial matrix near proximity to the mitochondrial respiratory chain, which serves as the primary endogenous source for superoxide. Three isoenzymes of SOD have been identified, though are related in functionality, vary widely in homology ranging from 17-23% between SOD1 which contains Cu and Zn ions as a homodimer existing in the cytoplasm where two 16 kDa subunits are linked by two cysteines forming an intra-subunit disulphide bridge and SOD3 (EC-SOD) which is a homotetramer of 30 kDa existing only in the extracellular space and can be distinguished by its heparin binding capacity which also contains Cu and Zn ions . However, SOD2's homology is preserved from species to species ranging from 87-90% homology match among rat and mouse SOD2. The vast majority oxidative stress within cells originates from the mitochondrion which is where SOD2 plays a key role in maintaining the structure and function of enzymes and their functionality and in particular, providing a defense mechanism for those enzymes which are susceptible to inactivation by superoxide. In homo sapiens, the SOD2 is encoded by the SOD2 gene residing on chromosome 6 and mutations associated with the SOD2 gene have been indicative of premature aging, sporadic motor neuron disease, idiopathic cardiomyopath (IDC) and cancer.
