NADH dehydrogenase (also known as cytochrome c reductase or NADH oxidoreductase) is another important respiratory chain linked mitochondrial enzyme that catalyzes transdehydrogenations between various pyridine nucleotides. This enzyme is known to be inactivated by oxidative stress. Hydroxyl radicals and superoxide anions were also shown to inactivate this enzyme by 60 and 40% respectively in the mitochondrial fraction. NADH dehydrogenase is measured at 340 nm in the presence of NADH and cytochrome C. The rotenone sensitive dehydrogenase activity (NADH oxidase) is calculated by subtracting rotenone insensitive NADH dehydrogenase activity in presence of rotenone from the total activity measured in the absence of rotenone. The assay procedure uses 1-ml assay reaction volume, and requires quartz cuvettes of 1 ml capacity and a spectrophotometer with kinetic function and equipped with a temperature control capable of maintaining 37°C.
