siRNAs are small interfering RNA or also sometimes referred to as short interfering RNA or silencing RNA are a class of double-stranded RNA molecules which are among the most abundantly utilized RNA Interference (RNAi) tools for inducing short-term silencing of protein coding genes. They operate within the RNAi pathway and are similar in length to miRNA (20-25 base pairs). Cells that receive transfection of synthetic oligonucleotides well allow for the siRNA to induce gene knockdown. Experimentation for transient silencing, which is possible with siRNA, endure a shorter duration ranging from 2-5 days. siRNA can also be utilized for the knockdown of proteins that are non-coding genes including long noncoding RNAs (lncRNA). siRNAs encompass two strands: an antisense guide strand and a passenger sense strand which forming a duplex of 19 to 25 bp in length. Double stranded siRNAs are often transfected into a cell where the guide strand is loaded into RRISC. This activated protein and nucleic acid complex may then allow for gene silencing by binding to a single target mRNA sequence due to its cleavage and degradation. Polymer based transfection reagents such as Redoxica's DINOFECT™ Transfection Reagent allow for the delivery of siRNA into the cell. Delivery can also occur via electroporation though this may cause in increase in cell death or by addition of chemical modifications to the duplex to assist in delivery though this method may trigger antiviral response in some cells.
