Succinate dehydrogenase is an iron-sulfur cluster protein and is consisted of two sub-units, one 70 kd and the other 27 kd and is embedded in the inner mitochondrial membrane. The electron acceptor of this important mitochondrial enzyme of the TCA cycle is the FAD, which is converted to FADH2 by succinate dehydrogense using two electrons from succinate and forming the fumarate. In the assay we employ K3Fe(CN)6 as the artificial electron acceptor instead of FAD. The rate of decrease in the concentration of K3Fe(CN)6 is monitored at 420 nm at 37°C. A blank reaction rate without succinate is always recorded for specificity. In the assay one mole of succinate reduces 2 moles of K3Fe(CN)6. The rate of reaction is calculated as nmoles of K3Fe(CN)6 reduced per minute at 37°C at pH 7.4 using molar extinction coefficient of K3Fe(CN)6.
