ZOLLY Isolation Reagent™ is a potent, monophasic, chaotropic phenol solution utilized for the simultaneous isolation and extraction of intact, undegraded biological molecules including DNA, RNA and protein from a wide variety of cells and tissues including cultured cell, human, animal and plant tissue, blood, yeast, viral, bacterial, and other challenging sample types. ZOLLY Isolation Reagent™ is based on Piotr Chomczyński and Sacchi's 1987 publication pertaining to guanidinium thiocyanate-phenol-chloroform based extraction, though significant enhancements were made in developing the final product. ZOLLY Isolation Reagent™ allows for the multi-layer precipitation of biological molecules from a single sample with better yields compared to other methods of extraction including guanidine thiocyanate/cesium chloride, phenol extraction and TCA-acetone precipitation, but with significantly less effort expended, a shorter timeframe, under 60 minutes . While utilizing the ZOLLY Isolation Reagent™, chloroform is exchanged with 1-bromo-3-chloropropane which allows for the effective lysing cells and tissues, deprotination of RNA, and inactivation of endogenous nucleases in a single-step. By lysing directly in the reagent to disrupt and break down cells and cellular components, molecular integrity of samples during tissue homogenization is preserved due to its inhibition of RNase/DNase activity and prevention of degradation during purification. Upon homogenation with ZOLLY Isolation Reagent™, chloroform is added to allow for the separation of the homogenate into a clear upper aqueous layer containing RNA (ranging from 0.1-15 kb in length), an interphase and a red, lower organic layer which contains DNA and proteins. Subsequently, isopropanol is used to separate the aqueous layer of RNA, ethanol may be utilized to precipitate the DNA from the interphase organic layer and isopropanol may be used to precipitate protein from the phenol-ethanol supernatant.
The resulting RNA, DNA and protein should be washed to remove impurities and then resuspended for downstream applications. The purified biological molecules including DNA, total RNA, transcriptome RNA, micro RNA, or protein is suitable for a wide range of downstream applications including northern blotting, mRNA isolation, cloning, cDNA library construction, RNase / DNase protection asssay, nuclease protection assays, in vitro translation, reverse transcriptase PCR (RT-PCR), and real-time quantitative PCR (RT-qPCR). ZOLLY Isolation Reagent™ is not compatible with high throughput processes and can only be conducted manually. ZOLLY Isolation Reagent™ has the ability to recovery biological molecules both large amounts of tissue (up to 1 g) and large amounts of cells (1 x 10^7 cells) or small amounts of tissue (40 mg - 110 mg) and small amounts of cells (5 x 10^6) making it ideal for a wide range of applications including gene expression and other work associated with both large and small sample quantities.
